5-Hydroxytryptophan (5-HTP), a precursor of serotonin, is therapeutically used for several psychiatric disorders such as anxiety and depression in the clinic. However, severe side effects, including abnormal mental functions, behavioral disturbances and intolerance are associated with this treatment. 5-HTP-induced elevation of plasma and brain serotonin levels may affect blood-brain barrier (BBB) breakdown, edema formation and regional cerebral blood flow (CBF) disturbances. Breakdown of BBB to serum proteins leads to vasogenic brain edema formation and cellular injuries. However, 5-HTP-neurotoxicity is still not well known. In this investigations 5-HTP induced elevation of endogenous plasma and brain serotonin levels and its effect on BBB breakdown, edema formation neuronal injuries was examined in a rat model. Furthermore, potential role of oxidative stress and nitric oxide (NO) was evaluated. In addition, several neurochemical agents such as p-CPA (5-HT synthesis inhibitor) indomethacin (prostaglandin synthase inhibitor), diazepam (ant stress drug), cyproheptadine, ketanserin (5-HT2 receptor antagonists) and vinblastine (inhibitor of microtubule function) were examined on 5-HT neurotoxicity. Our observations suggest that 4 h after 5-HTP administrations, the endogenous serotonin levels increased by fourfold (150 mg/kg) in the plasma and brain associated with profound hyperthermia (+ 3.86 ± 0.24 °C, oxidative stress and NO upregulation. Breakdown of the BBB to Evans blue albumin (EBA) in 8 brain regions and to [131]Iodine in 14 brain regions was observed. The CBF exhibited marked reduction in all the brain regions examined. Brain edema and cellular injuries are present in the areas associated with BBB disruption. Drug treatments reduced the BBB breakdown, edema formation NO production and brain pathology. These observations are the first to point out that 5-HTP-neurotoxicity caused by BBB breakdown, edema formation and NO production is instrumental in causing adverse mental and behavioral abnormalities, not reported earlier.View chapterPurchase bookRead full chapterURL: https://www.sciencedirect.com/science/article/pii/S007477421930025X

The 5-HT metabolic pathway is initiated by Trp being hydroxylated to the intermediate 5-hydroxytryptophan (5-HTP), which is subsequently decarboxylated to become 5-HT. Tissue levels of 5-HTP are usually low since this substance is rapidly decaroxylated by the enzyme aromatic amino acid decarboxylase [for review see (21)]. Thus, the rate limiting step in the biosynthesis of 5-HT is the hydroxylation of Trp which is catalyzed by the enzyme tryptophan hydroxylase (TPH) (Figure 1). This enzyme is specific for 5-HT producing cells, however, it is present in two different isoforms, TPH1 and TPH2 [reviewed in (22, 23)].

The hyper-response aims to deflate or attack the hyperthreat by operating at the microlevel through “mesh-interventions” as well as at the macrolevel through realignment of great nation states and tribes.
In IPCC AR6 WGIII Parlence, middle actors can mediate a community scale change, which becomes a force multiplier for individual change. Supercharging individual change is what can lead to significant scale of impact through many and many types of mesh interventions. The scale of such mesh interventions will have a "trickle up" effect to affect and accelerate the actions of top down actors.
This would be truly empowering as the current agency of the individual at the grassroots level is ineffectual.
Stop Reset Go (SRG) s a simple but powerful meme that is designed to be used by anyone to effect transition. When we recognize that something is harmful and needs to change, SRG can be invoked as a simple rule for transition. The colors of the traffic light are used as a mnemonic aid. If there is a problem with a human process, then STOP. think of an alternative way of achieving the same goal that does not bring about the harm (RESET). When the alternative has been trialed, tested and proven to work without causing more progress traps, then find a way to scale and implement the solution (GO).
SRG therefore becomes a simple mesh intervention that can be applied at all scales and dimensions. Its iterative and recursive use can be tracked in the Indyweb and interventions can be modeled by AI assistants that can analyze for potential unintended consequences through connections outside the focus area of the designer, and not normally explored by the designers. This augers a truly circular design methodology of the lowest potential impact.

IPCC AR6 WGIII Chapter 5: demand, services and social aspects of mitigation identifies that up to 45% of mitigation can result from a demand-side socialization strategy and collective action mobilization. This gives us tremendous power of impact to mobilize people. The low hanging fruit can be identified by comprehensive, ongoing, deep, global conversations with the greatest diversity of actors with a common vision collectively searching for the social tipping points, leverage points and idling resources and scaling massively thru the Indyweb as a cosmolocal network (what's light we share, what's heavy we produce locally).
Climate scientist and realist Professor Kevin Anderson has argued for many years that demand side changes are the only solutions that can be implemented rapidly enough to peak emissions and drop emissions rapidly in the short term (next few years), buying time for reneewable energy solutions to scale globally.

An analysis of “friendly forces” via a “tribal discourse” activity found that although many of humanity’s smaller and less powerful tribes are engaged in minor operations against the hyperthreat, its most powerful tribes often abet the hyperthreat (figure 2). If humanity’s tribes could be united against the hyperthreat, the current balance of probabilities, which currently lie with a hyperthreat victory and a Hothouse Earth outcome, could be recast.
This is the key idea behind mobilizing an effective global, multi-stakeholder, bottom-up response. Minor operations implies an aggregate approach that has little impact, otherwise known colloquially as "tinkering at the edge". IPCC AR6, WGIII Chapter 5 articulates this same message and for the first time, outlines that demand side system changes can play a significant role in mitigation effectiveness against the hyperthreat. It must be collectively organized individual change that scales to community scales around the globe in order to have impact, leveraging what the IPCC call "middle actors".
An effective strategy must be very time sensitive to the short time window to peak emissions so must identify all leverage points, idling resources and social tipping points available to a global bottom-up mobilization.

Polypropylene containers are autoclavable. The recommended autoclave cycle for empty containers is 121°C at 15 psi for 20 minutes. Care must be taken to allow free air circulation into and out of vessels during the autoclave cycle, especially during the venting and cooling stages. If the container is not properly vented, collapse or implosion (sometimes confused with melting) can occur. When autoclaving bottles and carboys, the cap threads must be completely disengaged from the container; the cap can be set loosely over the mouth opening at a rogue angle to ensure the threads don’t inadvertently engage. Once the container is completely cooled, the cap can be aseptically tipped into place and tightened down.
This is virtually identical to pressure cooker conditions. Thus these tips likely apply equally to cooking. Moreover, I take this as evidence that is appropriate for cooking food in the pressure cooker, though it may warp under stresses (so stacking containers may cause warping).

They comment on one another's work using technologies such as VoiceThread and support one another through instant messaging. Critical Thinking: When using the Internet, students build the text they read, choosing which links to follow and which to ignore. The nonlinear nature of online reading helps support critical thinking. Students also learn to question the perspective and bias of online sources. Comprehension: Students learn important online reading skills, such as how to distinguish news articles from blog posts and editorials. They carefully read texts they encounter online to understand and evaluate different perspectives.
