Here, we investigate CAF heterogeneity in a novel three-dimensional co-culture system that recapitulates the in vivo symbiotic interactions of CAFs and cancer cells. Our study reveals two spatially separated, mutually exclusive, dynamic, and phenotypically distinct CAF subtypes, underscoring the stromal heterogeneity in PDA and providing an opportunity to develop agents that target specific CAF populations.
