However, statistical analyses were found to lack rigor and clarity, and the lack of complementary experiments in females was noted as a weakness. This manuscript offers valuable information on the combinatory effect of small molecules, CHIR99021 and A-485 (2C), during the reprogramming of mature cardiomyocytes into regenerative cardiac cells on stimulating cardiac cell regeneration. Although the study used several hESC lines and an in vivo model of myocardial injury to demonstrate the regenerative potential of cardiac cells, the manuscript is still incomplete as several concerns remain unanswered, including the lack of validation of the conclusions from scRNA-seq. It is still unclear how a small fraction of dedifferentiating cardiac cells can offer such broad effects on regeneration both in vitro and in vivo. If validated, this study might unlock potential therapeutic strategies for cardiac regeneration.

This valuable article represents a significant body of work that addresses some novel aspects of the biology of lung cancer, the overall influence of CHIP and its impacts on responses to therapy. While a high clonal hematopoiesis (CHIP) burden was previously linked with an inflammatory phenotype in other disease settings, the authors demonstrate with solid evidence that this is also true for lung cancer. CHIP is complex and more data will be required to substantiate more evidence with regard perhaps to specific mutations in certain situations and how this might influence therapy choices.

Controversially, the authors report that forebrain glutamatergic synapses do not contain postsynaptic "densities" (PSD), a defining feature of synapse structure identified in chemically-fixed and resin-embedded brain samples. The work questions a long-standing concept in neurobiology and is primarily of interest to specialists in synaptic structure and function.

A solid model of brain aging using an open source algorithm is used. While some modest adverse brain health characteristics were associated with current mHT use and older age at last use, the findings do not support a general neuroprotective effect of mHT nor severe adverse effects on the female brain. The approach using mcherry adds to understanding of the role and localization of titin in controlling stiffness of striated muscles and fine tuning contraction. The results demonstrate that the integration of titin into the sarcomere is tightly regulated, with its unexpected mobility aiding in the uniform distribution of titin post-cell fusion. Although the experimental approach is convincing, the work is very qualitative in its approaches, and the data needs rigorous statistical analysis. There is a need for some clarification concerning numbers of animals and control groups. Future studies will need more rigorous data analysis and interpretation.

The landmark significance of this manuscript is based on the mechanistic description of purine metabolism by Lactobacillus plantarum, which helps to alleviate hyperuricemia, which is a phenotype that underlies multiple disease symptoms. The evidence provided for L. plantarum's involvement in reducing hyperuricemia was exceptional, combining microbiomics, whole genomics, in vitro bacterial culture, gene knock-outs, and metabolomics. Collectively, the study shows a clear link between the gut microbiota and hyperuricemia, providing a pathway for modification to help alleviate this condition.
