We observe significant declines in both website visits and question volumes at Stack Overflow, particularly around topics where ChatGPT excels. By contrast, activity in Reddit communities shows no evidence of decline, suggesting the importance of social fabric as a buffer against the community-degrading effects of LLMs.
Conclusion: LLMs cover certain areas that were before solved by human brainpower on stack overflow, and that is measurable over a period of oct-2021 -> march 2023. Reddit does not show this decline and this is alledged due to social fabric on reddit.
I want to know if it is possible to extract claims from this paper automatically in a way that it integrates certain relations in a knowledge graph, utilizing a system that improves itself.

Since less alcohol is required for isopropanol precipitation, this is the preferred method for precipitating DNA from large volumes. In addition, isopropanol precipitation can be performed at room temperature, which minimizes co precipitation of salt that interferes with downstream applications

Based on yesterday's discussion at Dan Allosso's Book Club, we don't include defense spending into the consumer price index for calculating inflation or other market indicators. What other things (communal goods) aren't included into these measures, but which potentially should be to take into account the balance of governmental spending versus individual spending. It seems unfair that individual sectors, particularly those like defense contracting which are capitalistic in nature, but which are living on governmental rent extraction, should be free from the vagaries of inflation?
Throwing them into the basket may create broader stability for the broader system and act as a brake via feedback mechanisms which would push those corporations to work for the broader economic good, particularly when they're taking such a large piece of the overall pie.
Similarly how might we adjust corporate tax rates with respect to the level of inflation to prevent corporate price gouging during times of inflation which seems to be seen in the current 2023 economic climate. Workers have seen some small gains in salary since the pandemic, but inflationary pressures have dramatically eaten into these taking the gains and then some back into corporate coffers. The FED can increase interest rates to effect some change, but this doesn't change corporate price gouging in any way, tax or other policies will be necessary to do this.

Looking for books with wider margins for annotations and notes
https://www.reddit.com/r/books/comments/wue2ex/looking_for_books_with_wider_margins_for/
Not long after I posted this it had about 3 upvotes, including my automatic 1. It's now at 0, and there are several responses about not writing in books at all. It seems like this particular book community is morally opposed to writing in one's books! 🤣
Why though? There's a tremendously long tradition of writing in books, and probably more so when they were far more expensive! Now they're incredibly inexpensive commodities, so why should we be less inclined to write in them, particularly when there's reasonable evidence of the value of doing so?
I might understand not writing in library books as part of their value within the commons, but https://booktraces.org/ indicates that almost 12% or more of the books they've tracked prior to 1924 have some sort of mark, writing, or evidence that it was actively read.
Given what I know of the second hand markets, it's highly unlikely that my books (marked up or not) will ever be read by another person.
There's so much more to say here, but I just haven't the time today...

When gold was discovered in the Yukon, 100,000 people desperately tried to make it to a small patch of land in one of the most remote environments on the continent. Few made it all the way. The Klondike Gold Rush was many things: a media conspiracy, a ponzi scheme, a land grab. But above all, it was a humanitarian disaster that stretched over much of the Pacific Northwest.
“The truth is that all of the gold that was mined out of the Klondike was under Indigenous land. There was no treaty with any of the Indigenous peoples in the Yukon.”
“That land was stolen by the Canadian state and that gold was whisked away by private interests. The Federal Government only signed land claims with Indigenous peoples in the Yukon in the 1990s, but by that point, almost all the gold had been mined out of the ground.”
“The Klondike gold rush was a rolling disaster that captured tens of thousands of people. When the first European explorers came to the Americas, they came here looking for gold. In the 1890s, that lust for precious metals eventually led men to the farthest reaches of this continent.”
“Today, instead of 100,000 people descending on a small patch of land, you have large corporations digging treasures out of the ground. But the legacies these mining operations leave behind are just like what happened in the Klondike: workers with broken bodies, environmental destruction, the dispossession of Indigenous land, sexual violence. The gold rushes never stopped. They just morphed into something different.”
Canada is Fake
“Canada is not an accident or a work in progress or a thought experiment. I mean that Canada is a scam — a pyramid scheme, a ruse, a heist. Canada is a front. And it’s a front for a massive network of resource extraction companies, oil barons, and mining magnates.”

This pattern lies at the heart of the shell corporation we call “Canada,” and forms the logic of both domestic and international policy. The mining industry is the most egregious example. Over 75 percent of the world’s mining companies are based in Canada.

Extraction Empire
Globally, more than 75% of prospecting and mining companies on the planet are based in Canada. Seemingly impossible to conceive, the scale of these statistics naturally extends the logic of Canada’s historical legacy as state, nation, and now, as global resource empire.
Canada’s Indian Reserve System served, officially, as a strategy of Indigenous apartheid (preceding South African apartheid) and unofficially, as a policy of Indigenous genocide (preceding the Nazi concentration camps of World War II).

The Doctrine of Discovery
Since the Doctrine of Discovery was issued by a Papal Bull in 1493, Western Europeans have used this document as legal justification for the genocide, colonization, extraction, and profit from the theft of land and its resources in what they called the New World.

Salvinorin A was notsoluble in 100% propylene glycol to our surprise. However, solubility increased in propylene glycol by adding increasingconcentrations of DMSO. A 9:1 mixture ended up beingsuitable for our studies
I'm quite a novice in chemistry. I assume a 9:1 ratio would work with other solvents replacing the 9 parts propylene glycol. That is to say, given that salvinorin A was not soluble in propylene glycol, I'm assuming the propylene glycol plays little to no role.

Exceptionally, chlorophyll b was not found in any of the top phases. The lack of chlorophyll a and chlorophyll b in some ATPSs could be due to low extraction efficiencies or a change in the molecule structure by oxidation. Chlorophyll stability is affected by temperature, light irradiance, acids, bases and oxygen, causing the loss of its magnesium ion and/or phytol group40 and a change in colour to olive‐brown.41
Which one it is should be possible to determine by running multiple washes and visually assess how much pigmentation is removed. The idea is to dissolve acetone plant extract, add PEG-400, let the acetone evaporate, pour PEG-400 leaving sediment and/or crystals, then repeat. While I'd probably be doing this with salvia divinorum, any leaf should work for testing chlorophyll washing.
-
Chlorophyll a was recovered by PEG400‐Ch DHp (50.6 ± 2.3%), but it was not detected in the other two systems at 660 nm [Fig. 2(b)].
They seem to be saying that PEG dissolved 50% of the chlorophyll? I also don't know what what Ch or DHp means. I'm somewhat outside my wheelhouse.
Assuming PEG-400 does not dissolve salvinorin A, then it may be an excellent option for purification of salvinorin A.

SA was soluble in 25% dimethyl sulfoxide (DMSO)/75% polyethylene glycol 400 (PEG-400) at 4,000 μg/mL and was stable for 6 days, as determined by LC-MS/MS, with only trace amounts of the hydrolysis product salvinorin B (SB) forming.
As with DMSO and propylene glycol, I suspect that the DMSO is doing the dissolving. Given that it appears to only take 10% DMSO to dissolve salvinorin, naturally the 25% works. Unfortunately, I see no mention of whether salvinorin A is soluble in pure PEG-400. I suspect it is not.

4. Allow acetone to evaporate in a dark, well-ventilated area. You will be left with a bright green powder – about 1g per 100g of leaf. This powder is about 2 parts chlorophyll to 1 part salvinorin A.
This is incredibly useful information.
I noticed my extract had some bitter taste and some earthy notes. I don't know what salvinorin and chlorophyll taste like, but I assume the taste comes from other impurities. However, I extracted for many minutes (10 or so); next time, I should do only the 3 minutes suggested here (3 times 1 minute).

In this study, we have quantitated yields of low copy and single copy number plasmid DNAs after growth of cells in four widely used broths (SB, SOC, TB, and 2xYT) and compared results to those obtained with LB, the most common E. coli cell growth medium
TB (terrific broth) consistently generated the greatest amount of plasmid DNA, in agreement with its ability to produce higher cell titers. The superiority of TB was primarily due to its high levels of yeast extract (24 g/L) and was independent of glycerol, a unique component of this broth. Interestingly, simply preparing LB with similarly high levels of yeast extract (LB24 broth) resulted in plasmid yields that were equivalent to those of TB.

What rationale is important to include in a brief? This is probably the most difficult aspect of the case to determine. Remember that everything that is discussed may have been relevant to the judge, but it is not necessarily relevant to the rationale of the decision. The goal is to remind yourself of the basic reasoning that the court used to come to its decision and the key factors that made the decision favor one side or the other.
